Version 2.5 includes support for Digital Audio and MIDI via the 30-pin dock connector

You need to go talk to IK Multimedia about this one...they aren't any more forthcoming with info than Alesis...I saw a recent post over there saying beginning 2012, however on past I'll believe it when I see it!

Some guy showcasing the Alesis AmpDock stated that it works with Amplitube..you might be better off just buying the iRig interface.

App wise, Ampkit is much better than Amplitube in my opinion.

Amplitube has some nice features (song tempo change and looping to learn phrases for instance) which are incredibly useful. Any news on when/if support will be coming?
